when it comes to secreted proteins, cell-free systems are an important and convenient tool that enables protein expression at an industrial scale. cell-free translation and transcription systems can be used to perform straightforward protein production as well as to study protein-protein interactions as well as protein-rna interactions. these complex processes are extremely difficult to study in living cells. here, we present and review the cell-free expression of the classical secreted marker protein for recombinant protein production, human serum albumin (hsa), as well as the secreted recombinant protein, the human-mouse chimeric antibody cetuximab. hsa is a well-established model protein in recombinant protein production, and cetuximab is a very promising anti-cancer antibody that is under preclinical and clinical development. the cetuximab cell-free expression system is a suitable model for the development of novel cell-free protein production platforms. the studies presented here show that the cell-free system is suitable to synthesize and secrete large amounts of hsa as well as cetuximab, and that both proteins are efficiently secreted. the study demonstrates that the cell-free system can be utilized to perform quality control as well as to obtain analytical information on the quality of the recombinant protein. the cell-free protein synthesis system also enables the production of multi-subunit complexes, such as antibody-antigen complexes, which cannot be produced in living cells.
