Pyruvate dehydrogenase kinase (PDK) is an important regulator of the TCA cycle. However, the role of PDK under hypoxia is not known yet. Therefore, in this study we examined the role of the PDK in striatal metabolism under hypoxia. Hypoxia (1% O2) caused a rapid decrease in glucose consumption and lactate production in striatal slices followed by a decrease in the ATP level. Incubation of slices with dichloroacetate, a pyruvate dehydrogenase kinase inhibitor, significantly reversed the effects of hypoxia. These data suggested that pyruvate dehydrogenase kinase may play an important role in controlling metabolism in the striatum during hypoxia. The lower production of ATP by PDK inhibition may result from the decreased pHi, as the metabolic fuel Pi is phosphorylated by the mitochondria, leading to an increase in pHi due to the decreased levels of ATP and high concentrations of Pi. To test this hypothesis, pHi was measured using a fluorescent dye, BCECF. Hypoxia caused a decrease in pHi in striatal slices which was partially recovered in the presence of dichloroacetate. These data suggest that PDK plays an important role in the maintenance of glycolysis during hypoxia, and a possible role for PDK inhibition to restore striatal pHi during ischemia is suggested
